Sample Preparation

NOTE: This protocol is a general guideline only, always follow the protocol at your institution Inspect: Check that the port and tubing are secure and in place. Check the integrity of the spike port and caps. Do not use if caps are not in place. If the bag is damaged or opened, DO NOT use it.

Label: Label all containers (bags and storage cassettes) according to your institutional protocols. There is a self-adhesive lot number label to which you may adhere a larger label.

Observe Labelling precautions! Caution: Do not write directly on the Cryostore Bag.

If you need to write on the outside of the bag, please use an Origen Ovewrwrap bag (see below), and write on that.

A label pocket has been provided on all Cryostore freezing bags. You may place a label inside this pocket, attach a label to, or write on this area. If a label or samples are to be stored in the pocket, it may be spot-sealed partially closed with your standard sealer. We recommend that you do not seal the entire edge to prevent rupturing due to potential liquid nitrogen infiltration.

WARNING: Complete sealing of label pocket may cause the label pocket to burst on re-warming.


Fluid Transfer: Cool the bags, cassettes, and any accessory equipment to be used to just above freezing for at least 30 minutes before using. The CryoStore bags have female luers attached to the extension lines and fluids can be introduced by accessing these ports.

Remove remaining air from the bag before sealing.

DMSO is potentially toxic to cells at temperatures above freezing. We recommend adding cryoprotectants to pre - chilled substrates and placing into a freezer immediately after mixing cell media.

Use a laminar flow hood, for an aseptic transfer technique.

Origen Overwrap bags

The OriGen O-Wrap™ Overwrap Bag is strongly recommended as way to ensure that samples and labels are not separated from the parent bag. Put everything, bag, labels and samples in the overwrap, and seal it closed. It also provides a backup safety system in the event that the inner bag is damaged, possibly allowing the sample to be recovered, and ensuring that the storage tank is not contaminated. The O-Wrap bag features

The O-Wrap bag is made of EVO copolymer, a custom formulation developed specifically for direct immersion in Liquid Nitrogen (LN). It remains flexible in LN, and is therefore ideally suited as a safety barrier for containing other freezing bags. The freezing bag is both very elastic and has a high puncture resistance, making it an ideal for cryogenic storage and shipment.

The O-wrap bag has greater than 90% optical transmission, making inspection of the contents easy, even when frozen.

The O-wrap bag is easily sealed with standard RF or inexpensive impulse heat seal equipment. Only a low temperature setting is required, and the resulting seal is clear, making inspection of the seal simple.

Because the O-Wrap bag is not used to contact other products, it can be labeled with adhesive labels or direct writing on the EVO bag.
